Hyphae are long tubular structures resembling garden hoses. However, all of these manifestations are formed of the same basic fungal unit, the hypha (plural: hyphae ). In 1-2 days they should look like obvious pins. This is what my GTs look like just as they are developing hyphal knots. There are a few areas where the mycelium seems to be clumping together. After approximately 120 h smooth undifferentiated primordia (23 mm diameter) were observed, some of which developed into elongated differentiated primordial. You will see hyphal knots prior to primordia formation. Our studies show that A mating type genes control all major differentiation processes in Coprinus, but whether developmental processes can proceed depends on the genetic background of the strain. Fungi can appear to be marvelously complex things, producing mushrooms, intricate coral-like structures and large brackets on trees. These cords thickened to form fluffy 0.51 mm diameter hyphal knots which developed into fluffy undifferentiated primordia (12 mm diameter) by approximately 95 h after venting. On artificial medium and on a 12 h light/12 h dark regime, A-activated transformants of one distinct monokaryon (218) formed fruit-body primordia that were arrested in development before karyogamy. Tmethyl Hyphal knotting close-up, inside a monotub (one day before fruiting) - Tmethyl Hyphal knots, primordia/pins - k00laid An expensive shot of Hyphal knots and Primordia (and some pins) growing on a 'cash casing' - RogerRabbit Pins growing in a wine glass - Tmethtyl A gorgeous set of pins - PrimalSoup An interestingly shaped pin set. Continuous incubation under blue light inhibited formation of hyphal knots, sclerotia and chlamydospores in both competent monokaryons and in A-activated transformants. Five of six monokaryons tested following transformation with A genes showed induced production of hyphal knots and sclerotia in the dark, and at least three strains showed enhanced chlamydospore production in the dark. Production of oidia in such A-activated transformants is repressed in the dark and induced by blue light. Hypha, hyphae individual cells of mycelium. That's why it is the most important phase in mushroom cultivation each grower waiting for it. The mushroom growth cycle is really cool if you look into it. Pinheads or primordia are knots of mycelium that proceed their development into small mushrooms. Primordial growth is the stage between hyphal knot formation and pinning. I see a lot of people refer to primordia as pins or knots. Monokaryons show various reactions upon transformation with heterologous A mating type genes. I see a lot of people worried when they see primordia formation. Some strains also produce large, inflated single cells (chlamydospores) at the agar/air interface, and hyphal aggregates (hyphal knots) that can develop into sclerotia. Monokaryons of Coprinus cinereus constitutively form small spores (oidia) in the aerial mycelium. RNA-seq revealed that some genes reported previously to be involved in formation of hyphal knots and primordia, cyclopropane-fatty-acyl-phospholipid synthases cfs1-3, galectins cgl1-3, and hydrophobins hyd1-3 were downregulated in the CcNsdD1/NsdD2-RNAi transformant compared to the mock transformant.
